Abstract

Aripiprazole adalah obat antipsikotik generasi kedua yang biasa digunakan untuk mengobati skizofrenia. Aripiprazole dikenal sebagai "penstabil sistem dopamin" dengan mekanisme aksi yang memberikan agonisme parsial dengan afinitas tinggi pada dopamin D2 dan serotonin-5-HT1A reseptor serta antagonisme pada serotonin-5-HT2A reseptor dan menunjukkan bukti kemanjuran klinis yang baik dengan profil keamanan dan tolerabilitas yang menguntungkan pada pasien dengan skizofrenia. Mekanisme kerja dari aripiprazole dianggap memungkinkan obat tersebut untuk memblokir jalur akibat kelebihan dopamin yang dapat menyebabkan psikosis, sementara pada saat yang sama aripiprazole dapat merangsang daerah otak untuk melakukan pengurangan terhadap dopamin sehingga dapat mengurangi efek samping. Karena insiden efek samping yang lebih rendah, aripiprazole sering dipilih sebagai pengobatan lini pertama untuk episode pertama psikosis.
